Observing cell physiological activity methods

Principle of experimental method
White blood cells are units that can travel in the body's defense system. It is divided into three types: granulocyte cell line, monocyte cell line and lymphoid line. They have many physiological functions, such as migration, deformation movement, chemotaxis, and phagocytosis of foreign bodies. In leukocytes, phagocytic activity of granulocytes and monocytes is strong, so these two types of cells are called phagocytic cells. Monocytes gradually evolve into macrophages after they enter the tissue. Phagocytic cells mainly rely on phagocytosis to treat foreign bodies. The phagocytic cells first migrate to the foreign body due to chemotaxis, and then extend the pseudopod to surround the foreign body, and endocytosis forms a phagocytic bubble to swallow the foreign body into the cell, and then the lysosome merges with the phagocytic bubble to digest the foreign body.

Experimental procedure
1. Two days before the experiment, the mice were intraperitoneally injected with 6% starch broth 0.5~1 ml (containing 0.3% trypan blue, labeled) to stimulate the peritoneal cavity to produce more macrophages (this step is performed by the teacher) Completed before the experiment).
2. During the experiment, one mouse in each group was treated with the above-mentioned treated mice, and 0.5% of the chicken blood suspension was intraperitoneally injected. After the injection, the abdomen of the mice was gently licked to make the red blood cell suspension evenly dispersed.
3. After 1 hour, the mice were sacrificed by cervical dislocation, and the abdominal wall of the mice was quickly dissected. 0.5 ml~1 ml of normal saline was injected into the abdominal cavity, and the physiological saline was mixed with the peritoneal fluid by gently stirring with a toothpick.
4. Use a syringe to draw the peritoneal fluid, drip, and then cover the cover.
5. Microscopic examination, the results of the high-magnification picture record.
