In fact, avian flu, like human flu, has never gone, and it often happens in flocks. And its horrible thing is not that it always exists, but because it "continuously mutates" and "never infects people, to passers-by, people pass people."

Influenza viruses can be classified into three types: A (A), B (B), and C (C). Among them, influenza A can be divided into H × N × 135 subtypes according to the characteristics of influenza virus, and H7N9 subtype avian influenza virus is one of them. Avian influenza A viruses can infect humans, pigs, horses, otters and marine mammals in addition to infected birds. The avian influenza virus subtypes that can infect humans are H5N1, H9N2, H7N7, H7N2, and H7N3. This report is a human infection with H7N9 avian influenza virus.

The avian influenza A virus particles are pleomorphic, wherein the spherical diameter is 80-120 nm, and there is a capsule. The genome is a segmented single strand negative strand RNA. According to the different antigenicity of the outer membrane hemagglutinin (H) and neuraminidase (N) protein, it can be divided into 16 H subtypes (H1 ~ H16) and 9 N subtypes (N1 ~ N9). The virus is a novel reassortant virus, the two internal gene fragments are derived from the avian H7N9 virus, and six are derived from the avian H9N2 virus, and there is no genetic fragment of the pig.

How to prevent and detect H7N9 virus in the population is not the purpose of this paper. This article mainly explains how to use laboratory equipment to isolate strains from diseased poultry tissue samples and collect avian influenza A virus from poultry house air.

Isolation of avian influenza A virus from diseased poultry tissue samples:
1. Take 200mg of diseased poultry lung tissue and put it into CK28 grinding tube.
2. Add TE buffer 1ml
3. Treat the Precellys24 biological sample homogenizer according to the parameter "6500rpm, 30s, 2cycles"
4. Centrifuge at 5000 rpm to remove the supernatant (Avian Influenza A virus is present in the supernatant and can be used for PCR detection)

Collecting avian influenza A virus from poultry house air:
1. Prepare Coriolis μ Air Biosampler and LTM Kit
2. Add TE buffer to the sample vial and connect the sample cup
3. Set the sampling rate of 200 L/min for 1 hr.
4. H7N9 virus is present in the sample cup liquid and can be used for PCR detection.

Note: Coriolis μ air biosampler must be equipped with LTM kit to effectively use Avian Influenza virus collection; the collection time is not less than 1hr; it will be inactivated because the virus is in solution for more than 30 minutes. It is recommended to carry out downstream PCR experiments as soon as possible. .
