The basic principle of the MassARRAY detection system is matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). The main feature is that the product after PCR amplification is added with a SNP sequence-specific extension primer and extends 1 base at the SNP site. The prepared sample analyte is then co-crystallized with the chip matrix, the crystal is placed in a vacuum tube of the mass spectrometer, and then excited by a strong nanosecond (10-9 s) laser, the nucleic acid molecules are desorbed and converted into singly charged ions. These singly charged ions fly in the accelerating electric field vacuum tube to the detector according to their mass-to-charge ratio. The smaller the ion mass, the faster it will arrive.
