
The research team used miRNA microfluidic chips (Lianchuan Biotech to provide technical services) and gene chip screening to analyze differentially expressed miRNAs and genes in bantam at the same developmental stage and at different developmental stages. At the same developmental stage, four miRNAs were differentially expressed in skeletal muscle of bantam (miR-1623, miR-181b, let-7b, and miR-128) compared to normal chicken. At different developmental stages, there are more differentially expressed miRNAs. Compared with 14-day-old embryos, 11 of the 7-week-old bantams were up-regulated and 18 were down-regulated, while in 7-week-old normal chickens, 7 were up-regulated and 9 down-regulated. In skeletal muscle,
22 genes were up-regulated and 33 down-regulated in 14-day-old embryos compared to
7-week-old bantam, whereas 65 genes were up-regulated compared to 7-week-old normal chickens. 108 expressions were down-regulated. Further, 34 differentially expressed miRNAs were divided into 18 categories according to overlapping seed regions and target sequences. Only the target of let-7b action is located in the 3' non-coding region of the GHR gene and can inhibit its expression. Three major signaling pathways were used to regulate skeletal muscle growth and fat deposition in chickens by KEGG Pathway analysis and qPCR experiments. These pathways are all affected by the GHR regulated by let-7b. The study also found that the Suppression Of the Cytokine Signaling 3 (SOCS3) gene is involved in the adipocytokine signaling pathway. *Lin SM, Li HM, Mu HP, et al .
